操作指南:
BD采用*利技術(shù),從富含胞外基質(zhì)蛋白的EHS小鼠腫瘤中分離出BDMatrigel基底膜基質(zhì),其主要成分由層粘連蛋白,Ⅳ型膠原,巢蛋白,硫酸肝素糖蛋白等組成,還包含生長(zhǎng)因子和基質(zhì)金屬蛋白酶等。BDMatrigel基底膜基質(zhì)在室溫條件下,聚合形成具有生物學(xué)活性的三維基質(zhì),模擬體內(nèi)細(xì)胞基底膜的結(jié)構(gòu)、組成、物理特性和功能,有利于體外細(xì)胞的培養(yǎng)和分化,以及對(duì)細(xì)胞形態(tài)、生化功能、遷移、侵染和基因表達(dá)的研究。
BDMatrigel基底膜基質(zhì)形成的三維培養(yǎng)基質(zhì),可促進(jìn)上皮細(xì)胞、肝細(xì)胞、Sertoli細(xì)胞、黑色素瘤細(xì)胞、血管內(nèi)皮細(xì)胞、甲狀腺細(xì)胞及毛囊細(xì)胞等的貼壁與分化。同時(shí),Matrigel還能影響乳腺上皮細(xì)胞的蛋白表達(dá),支持外周神經(jīng)的新生和牛輸卵管上皮細(xì)胞的分化。高濃度的Matrigel適用于研究體內(nèi)血管生成和腫瘤細(xì)胞遷移及腫瘤模型的建立等。
BD基質(zhì)膠/基質(zhì)膜cornning354248354230特性:
Matrigel基質(zhì)會(huì)有色差變化(淡黃色到深紅色),是由于酚紅和碳酸氫鹽與CO2作用引起的,但是與5%CO2平衡后色差即會(huì)減少。凍融后,輕輕搖晃試劑瓶使Matrigel分散均勻。所有操作均需在無(wú)菌環(huán)境下進(jìn)行,試劑瓶瓶蓋可用70%乙醇擦拭,并自然干燥。應(yīng)使用預(yù)冷的移液器以保證Matrigel呈勻漿狀。
細(xì)胞可在0.5mm厚度的Matrigel基質(zhì)層表面生長(zhǎng),也可在1mm厚度的Matrigel三維基質(zhì)內(nèi)生長(zhǎng)。過(guò)度稀釋的Matrigel會(huì)形成非膠質(zhì)的蛋白層,可以用于細(xì)胞貼壁,但不能用于細(xì)胞的分化研究。
注意:可將凍融后的Matrigel分裝在多個(gè)小管,所有分裝均需用預(yù)冷的凍存管,迅速冷凍并保存,避免多次凍融。
Matrigel在22-35℃溫度環(huán)境下快速成膠,因此溶解時(shí)在4℃冰上過(guò)夜凍融(4度時(shí)會(huì)隨著溫度的上升部分成膠)。所有用品在使用前需置于冰浴,必須使用預(yù)冷的移液管、吸頭及小管操作Matrigel。成膠后的Matrigel可以在4℃24-48小時(shí)后重新呈液態(tài)。
推薦包被與成膠方法:
注意:為了保證Matrigel基質(zhì)膜的成膠性能與穩(wěn)定性,稀釋濃度不應(yīng)低于1:3,可用無(wú)血清培養(yǎng)基稀釋,Matrigel成膠后立即使用。
薄膠成膠方法:
1.凍融后,用預(yù)冷的移液槍頭混勻Matrigel基質(zhì)成勻漿狀。
2.將需要使用的培養(yǎng)板置于冰上,加入濃度為50μL/cm2生長(zhǎng)面積的Matrigel基質(zhì)。
3.在37℃放置30分鐘,即可使用。
厚膠成膠方法:
1.凍融后,用預(yù)冷的移液槍頭混勻Matrigel基質(zhì)成勻漿狀。
2.將需要使用的培養(yǎng)板置于冰浴,將培養(yǎng)的細(xì)胞與Matrigel基質(zhì)混合,用移液槍頭使其懸浮于基質(zhì)中。加入濃度為150-200μL/cm2生長(zhǎng)面積的Matrigel基質(zhì)。
3.在37℃放置30分鐘,可成膠。可以加入細(xì)胞培養(yǎng)的基質(zhì),也可使細(xì)胞直接生長(zhǎng)在膠表面。
薄層包被方法:
1.凍融后,用預(yù)冷的移液槍頭混勻Matrigel基質(zhì)成勻漿狀。
2.根據(jù)使用需要,采用無(wú)血清培養(yǎng)基稀釋Matrigel基質(zhì)。根據(jù)實(shí)驗(yàn)需要確定*包被濃度。
3.將稀釋的Matrigel基質(zhì)包被于所需的培養(yǎng)器皿中,包被量至少覆蓋整個(gè)器皿的生長(zhǎng)表面。室溫下孵育1小時(shí)。
4.去除未結(jié)合的Matrigel,用無(wú)血清培養(yǎng)基輕輕地沖洗。
